Activation of neural cholecystokinin-1 receptors induces relaxation of the isolated rat duodenum which is reduced by nitric oxide synthase inhibitors BJMBR
Martins,S.R.; de Oliveira,R.B.; Ballejo,G..
Cholecystokinin (CCK) influences gastrointestinal motility, by acting on central and peripheral receptors. The aim of the present study was to determine whether CCK has any effect on isolated duodenum longitudinal muscle activity and to characterize the mechanisms involved. Isolated segments of the rat proximal duodenum were mounted for the recording of isometric contractions of longitudinal muscle in the presence of atropine and guanethidine. CCK-8S (EC50: 39; 95% CI: 4.1-152 nM) and cerulein (EC50: 58; 95% CI: 18-281 nM) induced a concentration-dependent and tetrodotoxin-sensitive relaxation. Nomeganitro-L-arginine (L-NOARG) reduced CCK-8S- and cerulein-induced relaxation (IC50: 5.2; 95% CI: 2.5-18 µM) in a concentration-dependent manner. The magnitude...

DENSITY AND MORPHOMETRY OF MYENTERIC NEURONS OF THE ILEUM OF RATS SUBJECTED TO CHRONIC ALCOHOLISM International Journal of Morphology
dos Santos Pereira,Marli Aparecida; Molinari,Sonia Lucy; de Sousa,Fernando Carlos; André,Oriana Elaine; de Miranda Neto,Marcílio Hubner.
This work verified the changes caused by ethanol ingestion on the myenteric neurons of the ileum of rats subjected to chronic alcoholism. Twenty rats (Rattus norvegicus) were divided in control (GC) and alcoholic (GA) groups during 210 days. The animals from group GA received sugar-cane brandy diluted to 30 GL and those from group GC received water. Both groups were fed with rodent chow ad libitum. The ileum was prepared as whole-mounts for neuronal staining according to Barbosa (1978) and Gabella (1979). The neurons were counted and the cell bodies were measured. The statistical analysis revealed a significant decrease in the number of large neurons in GA compared with GC, while there was a significant increase in the number of small neurons in GA. There...

SOUZA,NOEMI D. DE; BELIN,BRUNA S.; MASSOCATTO,CRISTINA L.; ARAÚJO,SILVANA M. DE; SANT’ANA,DÉBORA M.G.; ARAÚJO,EDUARDO J.A.; P. FILHO,PHILENO; NIHEI,OSCAR K.; MOREIRA,NEIDE M..
Abstract: We investigated the effects of acetylsalicylic acid (ASA) on the total myenteric neuronal population in the descending colon in Trypanosoma cruzi-infected mice. Thirty-five male Swiss mice, 60 days old, were divided into a control group (C group), control group treated with ASA (CA group), infected group (I group), and infected group treated with ASA (IA group). A total of 1300 trypomastigotes of the Y strain of T. cruzi were intraperitoneally inoculated in the IA and I groups. The CA and IA groups were treated with ASA intraperitoneally. At 75 days post-infection (dpi), all of the animals were sacrificed. Neurons in the colon were stained with Giemsa, quantified, and measured. No difference in the course of infection was observed between the IA...

Effect of the ascorbic acid treatment on the NADHd-positive myenteric neurons of diabetic rats proximal colon BABT
Zanoni,Jacqueline Nelisis; Pereira,Renata Virginia Fernandes; Freitas,Priscila de.
The aim of this work was to study the effect of the ascorbic acid on the myenteric neurons of diabetic rats proximal colon. Fifteen rats (90 days old) were divided into three groups: control, untreated diabetic and treated diabetic with ascorbic acid (DA). After 120 days of daily treatment with ascorbic acid, the intestinal segments were submitted to the NADH-diaphorase (NADHd) histochemistry technique to expose the myenteric neurons. The group DA showed a higher neuronal density (33.4 %) when compared to the untreated diabetic animals (p < 0.05). Cellular body area of neurons was significantly larger in group DA (17.3 %) when compared to the untreated diabetics (p < 0.05). It could be concluded that the ascorbic acid promoted a neuroprotective...

Effects of the ascorbic acid supplementation on NADH-diaphorase myenteric neurons in the duodenum of diabetic rats Biocell
Pereira,Marli Aparecida Dos Santos; Bagatin,Maria Claúdia; Zanoni,Jacqueline Nelisis.
We assessed the ascorbic acid (AA) supplementation on the myenteric neurons in the duodenum of rats. Fifteen rats with 90 days of age were divided into three groups: control (C), diabetics (D) and ascorbic acid treated diabetics (DA). After 120 days of daily treatment with AA, the duodenum was submitted to the NADH-diaphorase (NADH-d) histochemical technique, which allowed us to evaluate the neuronal density in an area of 8.96 mm² for each duodenum, and also to measure the cellular profile area of 500 neurons per group. The supplementation promoted an increase on AA levels. The neuronal density (p <0.05) was higher in the group DA when compared to group D. There were no significant differences in the neuronal areas, when we compared groups C (204 ±...

Food restriction beginning at lactation interferes with the cellular dynamics of the mucosa and colonic myenteric innervation in adult rats Anais da ABC (AABC)
SCHOFFEN,JOÃO PAULO F.; VICENTINI,FERNANDO A.; MARCELINO,CAROLINA G.; ARAÚJO,EDUARDO J.A.; PEDROSA,MARIA M.D.; NATALI,MARIA R.M..
The effects of food restriction (FR) on the morphoquantitative aspects of the wall and myenteric neurons of the proximal colon in adult rats were analysed. FR was imposed by duplication of the experimental brood size in relation to the control brood during lactation. The FR group received a 50% reduction of food from weaning until 90 days of age. Samples of the colon underwent histological processing to morphometrically analyze the crypts, muscularis mucosae, tunica mucosa, and muscularis externa. We determined the number of goblet cells and serotoninergic enteroendocrine cells, and morphoquantitatively studied the myenteric neuronal population. FR caused hypertrophy in the tunica mucosa, increase in crypt depth and in the muscular layer of the mucosa, a...

Pereira,Marli Aparecida dos Santos; Arantes,Márcia Alessandra; Molinari,Sonia Lucy; Zanoni,Jacqueline Nelisis; Miranda-Neto,Marcílio Hubner de.
The objective of our work was to verify, during an experimental period of 210 days, the possible effects of the ethanol on the morphology and density of the NADH-diaforase myenteric neurons in the jejunum of alcoholic rats. We used 10 animals (Rattus norvegicus) separated in 2 groups: the controls (n=5), that received a normal proteic diet (22%) and water ad libitum; the alcoholic, that received NUVILAB normal proteic chow (22%) and sugar cane brandy diluted at 30 Gay Lussac (30 v/v). The jejunum segment was collected and submitted to the histochemical technique to stain the nervous cells and, then, to the elaboration of membrane whole mounts. The observation through light microscopy showed that there are no expressive morphologic differences between the...

MORPHOMETRY AND QUANTIFICATION OF THE MYENTERIC NEURONS OF THE DUODENUM OF ADULT RATS FED WITH HYPOPROTEIC CHOW International Journal of Morphology
Marçal Natali,Maria Raquel; Hubner de Miranda-Neto,Marcílio; Orsi,Antônio Marcos.
Morphometric and quantitative analyses were carried out on the myenteric neurons of the duodenum, to study possible alterations resulting from a supply of low-protein level chow to adult Wistar rats. The animals were divided in two groups: nourished (N) fed with ration containing 22% protein for 210 days and the undernourished (D) fed with ration containing 8% protein from the 90th day of age on and during the next 120 days. Whole-mounts were stainedby the method of Giemsa and the histochemical reaction of NADH-diaphorase enzyme, to estimate the neuronal population, which revealed a greater neuronal density in undernourished rats, regardless of the technique employed. The morphometric studies of the cell body profiles of 500 neurons of each group indicated...

Morphoquantitative evaluation of the duodenal myenteric neuronal population in rats fed with hypoproteic ration Biocell
Marçal Natali,María Raquel; Molinari,Sonia Lucy; Valentini,Luiz Cristiano; Hubner de Miranda Neto,Marcílio.
The purpose of this work was to analyze the morphoquantitative behavior of the neurons of the myenteric plexus, as well as the morphometry of the duodenal wall, in adult rats fed with normoproteic (22%) and hypoproteic (8%) rations, killed at the age of 345 days. For neuronal assessments duodenal wholemounts stained with the Giemsa method were used, and for the evaluation of the duodenal wall routine histological processing and staining with Hematoxilin-Eosin were employed. The means of the number of neurons in 80 microscopic fields (12.72 mm2) and of the size of the neuronal cell bodies did not reveal statistically significant differences between the groups, but there was a greater incidence of large neurons in the protein restriction group (RP). The...
